New time-scale criteria for model simplification of bio-reaction systems

Background:Quasi-steady state approximation (QSSA) based on time-scale analysis is known to be an effectivemethod for simplifying metabolic reaction system, but the conventional analysis becomes time-consuming andtedious when the system is large. Although there are automatic methods, they are based on eigenvaluecalculations of the Jacobian matrix and on linear transformations, which have a high computation cost. A moreefficient estimation approach is necessary for complex systems.Results:This work derived new time-scale factor by focusing on the problem structure. By mathematicallyreasoning the balancing behavior of fast species, new time-scale criteria were derived with a simple expressionthat uses the Jacobian matrix directly.

Background:Beta-thalassemia is a common autosomal recessive hereditary disease in the Meditertanean, Asia and African areas. Over 600 mutations have been described in the beta-globin (HBB), of which more than 200 are associated with a beta-thalassemia phenotype.Results:We used two highly-specific mutation screening methods, mismatch-specific endonuclease and denaturing high-performance liquid chromatography, to identify mutations in the HBB gene. The sensitivity and specificity of these two methods were compared. We successfully distinguished mutations in the HBB gene by the mismatch-specific endonuclease method without need for further assay.
